Passage of Fucose-3h Label from the Golgi Apparatus into Dense and Multivesicular Bodies in the Duodenal Columnar Cells and Hepatocytes of the Rat
نویسندگان
چکیده
Fucose 3H is a stable sugar selectively taken up into glycoproteins (1, 2) . Since fucose residues are located at the end of carbohydrate side chains (3), the uptake of fucose indicates completion of the synthesis of these side chains and presumably of the glycoprotein molecules themselves . When duodenal columnar cells were examined after fucose1H injection into rats, the label was observed only in the Golgi apparatus at 2 min, but by 20 min after injection was also found at all cell surfaces (4) . Presumably a glycoprotein completed in the Golgi apparatus was rapidly transferred to the outer surface of the plasma membrane to be added to the "cell coat ." In the rest of the cell, some radioactivity was associated with vesicles, which may be the carriers of glycoprotein from Golgi apparatus to cell coat (4, 5) . It was also noted recently that some radioactivity was located in dense and multivesicular bodies (6, 7) . A similar finding has been reported in lymphocytes (8) . The present investigation is a systematic examination of this phenomenon in duodenal columnar cells and hepatocytes.

Isolation and characterization of multivesicular bodies from rat hepatocytes: an organelle distinct from secretory vesicles of the Golgi apparatus
Hepatocytes of estradiol-treated rats, which express many low density lipoprotein receptors, rapidly accumulate intravenously injected low density lipoprotein in multivesicular bodies (MVBs). We have isolated MVBs and Golgi apparatus fractions from livers of estradiol-treated rats. MVB fractions were composed mainly of large vesicles, approximately 0.55 micron diam, filled with remnantlike very...
متن کاملEvaluation of the Stress Effect on Expression of BEST-5 Gene in Gultured Rat Hepatocytes and Cloning of This Gene
Introduction:Liver has important roles in body metabolic regulation and for this reason hepatocytes are used worldwide.Investigations showed that isolation of hepatocytes causes activation of stress related genes. The aim of this study was to study the stress related expression of BEST-5 following hepatocytes isolation and culture. Method:The BEST-5 gene is cloned and analyzed for the first tim...
متن کاملHEPATOMA McARDLE-RH7777 CELLS HAVE THE SAME RESPONSE AS NORMAL RAT HEPATOCYTES TO BOTH DIBUTYRYL-cAMP AND ANTICALMODULINW-7
The effects of cAMP-analogue dibutyryl-cAMP and anticalmodulin W-7 were studied on de novo synthesis and secretion of lipids in cultures of hepatoma McArdle RH7777 cells and normal rat hepatocytes. Dibutyryl-cAMP and W -7 separately caused a significant decrease in the secretion of de novo synthesized triacyl [3H]glycerol in both cultures of McArdle cells and rat hepatocytes. The inhibito...
متن کاملFormation of Cell Coat Material for the Whole Surface of Columnar Cells in the Rat Small Intestine, as Visualized by Radioautography with L-fucose-3h
Some 30 years ago, Chambers proposed that the cell surface is covered by a thin amphous `cell coat" (1) . The evidence in support of this view was mostly indirect until the cell coat was demonstrated in numerous rat cells by light microscopy with the periodic acid-Schiff or colloidal iron procedure (2) and by electron microscopy with periodic acid-silver methenamine (3) or phosphotungstic acid ...
متن کاملMigration of Glycoprotein from Golgi Apparatus to Cell Coat in the Columnar Cells of the Duodenal Epithelium
A layer of carbohydrate material appears at the surface of a variety of mammalian cells (1) and is particularly abundant at free surfaces such as the striated border of the columnar cells of the intestine (2). In electron micrographs of intestinal cells, this material appears as a discrete coating covering the tips and sides of the microvilli (3). From the various histochemical tests applied (3...
